The wavelength knob on a spectrophotometer affects the particular wavelength of the light that is being allowed to shine on the sample in the cuvette. Different colors of light are expressed at these wavelengths. If you think of the light that you see shining off of a regular bulb, it appears to be white. However, when you put a prism in front of the beam of light, it breaks the light down into individual colors to form a spectrum. A spectrophotometer is a machine that does this and the wavelength knob determines what small portion of that spectrum will be allowed to shine through onto the sample. For example, if you had an unknown sample and were trying to determing the spectrum at which it absorbed the light, you would start with your knob at 350 nm. Then you would go up by increments of 10 or so on the knob and take the readings.
